A team of embryologists has uncovered the significant role that the expansion and contraction of embryos play during this critical phase.
Using advanced time-lapse video and high-resolution imaging, they observed that frozen embryos that don’t re-expand by 20-50% within the first couple of hours post-thawing tend to have a much lower chance of leading to a successful pregnancy. The Impact of Vitrification on Embryo Hatching
While earlier studies hinted that the freezing process (vitrification) could alter the embryo’s outer membrane (known as the zona pellucida), no one had previously examined its effects on embryos hatching on Day 5, or what we call blastocysts. In this study, blastocysts were photographed right after they were warmed and again after one and two hours. By comparing these volume changes with transfer outcomes, they were able to analyze the hatching ability of the embryos.
The results showed that the elasticity of the zona pellucida decreased significantly after vitrification. Interestingly, even with small holes from cell biopsies used for genetic testing, embryos experienced delays in hatching. Some were stuck for 6 to 48 hours, despite good volume re-expansion.
Findings on Assisted Hatching
On the flip side, when researchers created a larger opening in the zona pellucida, the warmed embryos managed to hatch completely within four hours. They also found that embryos that expanded in volume by at least 20% within the first hour or 50% after two hours had significantly higher clinical pregnancy rates. Dr. Smith remarked that this study suggests aggressive assisted hatching could be beneficial for certain embryos.
